谷胱甘肽S-转移酶M_3基因多态性与支气管哮喘易感性

摘要 目的探讨谷胱甘肽S-转移酶M3(GSTM3)内含子6是否存在AA、AB、BB基因多态性,并研究其与哮喘易感性的关系。方法抽取53名哮喘患儿(病例组)和44名正常儿童(对照组)外周静脉血2mL,提取DNA,并利用限制性内切酶MnlⅠ,聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析及基因测序等技术检测GSTM3是否存在AA、AB、BB3种基因型,如果存在,则用χ2检验比较病例组与对照组之间基因型频率。结果本实验未发现GSTM3存在基因多态性,聚丙烯酰胺凝胶电泳(PAGE)检测不到RFLP。结论所研究人群中GSTM3基因不存在多态性。 Objective To investigate the existence of the polymorphism AA, AB, BB which is located in intron 6 of Glutathione S-transferase M3 (GSTM3) gene and the association between the mutation and asthma. Methods The DNA was extracted from 2mL venous blood from 53 asthma subjects and 44 normal subjects. Polymorphism chain reaction-restriction fragment length polymorphism (PCR-RFLP), analysis restriction endonuclease Mnl Ⅰ and DNA sequencing were used to detect the genotypes of AA, AB, BB. If the mutation exist, x^2 analysis was used to test allele frequency and genotype frequency. Results The polymorphism in intron 6 in GSTM3 gene was not detected in this research and RFLP was not available in the polyacrylamide gel electrophoresis(PAGE). Conclusion RFLP in GSTM3 gene was not detected in our study.
