醒脑静粉针对实验性脑外伤的保护作用 被引量:11

摘要 目的研究醒脑静粉针(Xingnaojing powder,XNJP)对小鼠脑外伤致脑功能障碍及创伤性脑损伤后大鼠脑组织线粒体谷胱甘肽(GSH)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平的影响。方法采用悬垂试验及被动活动试验,观察自由落体脑挫裂伤模型静脉注射醒脑静后的神经功能及行为症状;分别采用黄嘌呤检测法、硫代巴比妥酸检测法及比色法测定给药4 h后大鼠脑组织线粒体GSH、SOD和MDA含量。结果脑外伤后静脉注射400、800 mg.kg-1醒脑静粉针,能显著提高脑外伤小鼠悬垂评分及延长悬垂时间和被动活动时间;800 mg.kg-1醒脑静粉针治疗4 h,脑组织线粒体SOD、GSH含量均高于模型组(P<0.01),而MDA含量均显著低于模型组(P<0.01)。结论醒脑静粉针对小鼠脑外伤造成的部分肌力丧失及共济运动部分失调有保护作用;能够减轻脑外伤造成的脑水肿损害;可以通过影响线粒体功能而减轻创伤性脑损伤后的继发性脑损害。 Objective To study the effect of Xingnaojing ( traditional Chinese medicines) powder (XNJP) on the brains dysfunction induced by trauma in mice and on the content levels of GSH, SOD and MDA of mitochondria in rats' brain tissues after the trauma. Methods Suspending experiments and passive activities experiments were applied to observe the protective effect of XNJP(i. v. ) The trauma model was established by free falling of the weight upon the heads. XNJP was injected intravenously immediately after the trauma. Then biochemical assays were applied to determine the content levels of GSH, SOD and MDA 4 hours after the trauma. Results Injection of XNJP of the dose of 400 mg. kg^-1 and 800 mg.kg^-1 (i. v. ) could significantly increase the performance marks of mice in the suspending experiments; prolong the length of suspending time and the time of passive activity. The content level of SOD and GSH in mitochondria in brain tissues were improved significantly compared to the model group (P 〈 0.01 ), while the content levels of MDA was obviously lower compared to the model group (P 〈 0.01 ). Conclusions XNJP has a protective effect on the partial muscle impotency and partial ataxia induced by brain trauma. Meanwhile it can relieve the brain edema induced by the trauma. XNJP works through imposing its efficacy on the mitochondria so that it can reduce the secondary brain damage induced by brain trauma. As a result, the prognosis is improved.
