摘要 目的探讨记录全细胞的大电导钙激活钾通道的技术方法。方法首先酶解急性分离大鼠海马神经元;利用全细胞膜片钳技术记录大电导钙激活钾电流。结果可记录到一系列快速的外向钾离子电流。结论所记录到的外向钾离子电流中,快速出现并很快减小的呈尖峰样的瞬变外向电流主要由大电导的钙激活钾电流组成。 Objective To explore a method for the whole currents record of a large-conductance Ca^2+ -activated K^+ currents in rat hippocampal CA1 neurons. Methods After rats hippocampal CA1 neurons were isolated by protease, the large-conductance Ca^2+ - activated K^+ currrents were recorded in rats hippocampal neurons by whole cell damping technique. Results A very strong outward currents appeared. Conclusion The outward currents are composed of transient outward currents with multiple compounds were recorded and sustained outward currents. And Ca^2+ -activated K^+ channels mediate a major part of the transient outward.
