酸性染料比色法测定槲寄生总生物碱的含量

Determination of the total alkaloids in ramulus visci by acid dye colorimetry

目的建立槲寄生总生物碱含量测定方法,比较不同产地槲寄生总生物碱的含量。方法以硫酸阿托品为对照品,利用酸性染料比色法,在420 nm处测定槲寄生总生物碱含量。结果在25-125μg/ml范围内线性关系良好(r=0.999 7),平均回收率为102.8%(RSD=2.5%),样品在2-6 h内测定稳定。结论该方法准确、可靠,可用于槲寄生总生物碱的含量测定;经测定云南、山西、内蒙、辽宁不同产地槲寄生总生物碱含量分别为0.84,0.92,1.10,3.80 mg/g。

Objective To establish the determination method of total alkaloids in ramulus visci （RV） and to compare the contents of total alkaloids in RV from different places. Methods The total alkaloids were tested at 420 nm with acid dye colorimetry in which atropine sulfate was used as standard reference. Results A good linear correlation was found in the range of 25 to 125 μg/ml（ r = 0. 999 7）. The average recovery was 102.8% （RSD= 2.5% ）. The sample was stable when it was determined during 2 - 6 h. Conclusion This method is accurate and credible. The contents of total alkaloids in RV from Yunnan, Shanxi, Neimeng, Liaoning are 0.84,0.92,1.10,3.80 mg/g.
