摘要
目的:探讨钙蛋白酶10基因多态性与原发性高血压遗传易感性的关系。方法:采用病例对照研究设计,以聚合酶链式反应-等位基因特异性扩增法(PCR-ASA),检测Calpain10基因SNP43G/A位点和SNP44T/C位点基因型,并进行与原发性高血压的关联分析。结果:在非糖尿病人群中,原发高血压患者和血压正常者的钙蛋白酶10基因SNP43GG基因型频率分别为70.59%和72.73%,差异无统计学意义,其SNP44TT基因型频率分别为80.88%和78.41%,差异亦无统计学意义;分析原发性高血压与血糖、真胰岛素、C肽及胰岛素抵抗指数HOMA-IR等的关系,高血压患者与正常人群各项临床指标之间的差异无统计学意义(P>0.05)。结论:钙蛋白酶10基因与昆明高原地区人群原发性高血压的发病无关,同时亦未发现原发性高血压与胰岛素抵抗的相关性。
Objective. To explore the hypothesis that Calpain10 gene contributes to the genetic susceptibility in hypertension in plateau population. Methods: In case oontrol study, PCR- ASA method was used to determine the distribution of allele and geno- type frequencies of SNP43 polymorphisrn (G/A) and SNP44 polymorphism (T/C) of calpain10 gene in 68 hypertensive patients and 88 normal oontrol subjects. Results: The frequency of "GG" genotype of the SNP43 in hypertensive patients is as same as that in the oontrol subjects; and so is the frequency of "TT" genotype of the SNP44. The variation of clinical metabolic parameter in hypertensive patients is also as same as that in the oontrol subjects. Conclusion: The study suggested that Calpain10 gene is not associated with hypertension in plateau population in Kunming; and the oorrelation between hypertension and insulin resistance is not found either.
