L-精氨酸对大鼠肾缺血再灌注损伤细胞凋亡的影响 被引量:5

摘要 目的探讨左旋精氨酸(L arginine,L arg)对大鼠肾缺血再灌注损伤(RIRI)细胞凋亡的影响。方法采用夹闭双侧肾动、静脉45min后松夹再灌的方法制作RIRI模型,测定血清肌酐(Scr)、丙二醛(MDA)、一氧化氮(NO)及尿NAG酶(UNAG)含量,观察肾组织形态学改变和肾皮质细胞凋亡率。结果缺血再灌注后6和24h,大鼠肾功能和肾小管上皮细胞明显受损,Scr、MDA和UNAG含量均明显高于假手术组(P<0.01),肾皮质细胞凋亡率明显增加。尾静脉注射L arg可明显改善RIRI大鼠肾功能,减轻肾损伤,并明显降低肾皮质细胞凋亡率(P<0.05)。结论L arg可通过增加体内NO水平,抑制肾小管上皮细胞凋亡对大鼠RIRI起到一定的保护作用。 Objective To investigate the effect of L-arginine on cell apoptosis caused by renal ischemia reperfusion injury (RIRI) in rats,Methods The rat model of RIRI was induced by bilateral clamping the renal artery and vein for 45 minutes followed by reperfusion. Serum creatinine (Ser) and urea N-Acety-P-beta Glucosaminidase (UNAG) were measured to evaluate renal function. Nitric oxide (NO), malondialdehyde (MDA) and renal histological changes were tested to observe the degree of renal injury, Flow cytometer was used to estimate the apoptosis rate of renal cortex cells. Results The contents of Set, MDA and UNAG significant increased in RIRI rats ( P 〈 0.01 ). and serum NO levels were also increased in RIRI rats, but there was no significantly difference, as compared with those in sham operation group; the apoptosis rate of cell in renal cortex was obviously increased in RIRI rats ( P 〈 0.01). L-arginine significantly decreased the contents of Ser, MDA, UNAG and L-arginine also could mitigated the apoptosis rate of renal cortex cells in early reperfusion and could attenuate the renal histological injury in RIRI rats. Conclusion L-arginine has protective effect against renal dysfunction in RIRI rats by increasing the levels of NO and by inhibiting the apoptosis of the renal cortex cells.
