摘要
目的:测定万应胶囊(胡黄连、黄连、儿茶等)中儿茶素、表儿茶素和盐酸小檗碱的含量。方法:采用反相高效液相色谱法,D iamonsidTM-C18柱,柱温为室温。儿茶素、表儿茶素的测定:以0.04 mol/L枸橼酸溶液-N,N-二甲基甲酰胺-四氢呋喃(40∶8∶2)为流动相,流速为1.0 mL/m in,检测波长280 nm;盐酸小檗碱的测定:以乙腈-水(52∶48)(含0.34%磷酸二氢钾和0.17%十二烷基硫酸钠)为流动相,流速为1.0 mL/m in,检测波长265 nm。结果:儿茶素平均回收率为100.9%,方法精密度(RSD)为2.52%(n=6);表儿茶素平均回收率为99.9%,方法精密度(RSD)为2.53%(n=6);盐酸小檗碱平均回收率为97.0%,方法精密度(RSD)为2.70%(n=6)。结论:该法可用于该制剂的质量控制。
AIM: To determine the content of ( + )-catechin and ( - )-epicatechin and berberine hydrochloride in Wangying Capsule (Rhizoma Picrorhizae, Rhizoma Coptidis, Catechu, etc. ). METHODS: A RP-HPLC method was established. The chromatographic column was Diamonsil-C18. The mobile phase was 0.04 mol/L citric acid-N, N-dimethylformamide-tetrahydrofuran (40:8:2) for determining ( + )-catechin and ( - )-epicatechin with the flow rate of 1.0 mL/min, and the detection wavelength of 280 nm. The mobile phase was acetonitrile-water (52 :48 ) (including 0.34% potassium dihydrogen phosphate and 0.17% sodium dodecyl sulfate) for determining berberine hydrochloride with the flow rate of 1.0 mL/min, and the detection wavelength of 265 nm. RESULTS : The average recoveries for ( + ) -catechin and ( - ) -epicatechin and berberine hydrochloride were 100.9% and 99.9% and 97.0%, precision of the method were 2.52% and 2.53% and 2.70% (RSD, n = 6 ), respectively. CONCLUSION: The method can be used for quantitative determination of the preparation.
