TRUGENE HIV-1基因型检测系统在HIV-1亚型分析中的应用

摘要 目的利用TRUGENE HIV-1基因型检测系统对我国部分地区HIV感染者的血浆样本进行耐药性检测的同时进行HIV-1的亚型分析。方法采用TRUGENE HIV-1试剂盒扩增靶HIV-1序列即pol基因中长度分别为297bp的蛋白酶基因片段和621bp的逆转录酶基因片段,采用OpenGene DNA测序系统和数据分析软件进行测序和序列分析,利用HIV Database网站提供的HIV基因亚型分析软件对所得到的靶序列进行亚型分析。结果对165份标本的靶序列进行扩增和测序全部成功,采用蛋白酶基因片断和反转录酶基因片断进行亚型分析的结果非常吻合,165份标本亚型的分类与分布情况提示目前各类感染人群中的HIV-1毒株的亚型分布特点呈现出多样化和复杂性。另外,发现了一例国内尚未报道过的CRF09-cpx HIV-1的感染。结论TRUGENE HIV-l基因型检测系统可以成功的应用于HIV-1的亚型分析。 Objective To detect 165 samples from different regions in China for drug resistance analysis using TRUGENE, at the same time get subtype information of the samples. Methods Amplify and sequencing target genes which are 297bp protease gene and 1680bp reverse transcriptase gene regions using TRUGENE HIVol genotyping kit and OpenGene DNA sequencing system, Determination of HIV-1 subtypes was done by means of BLAST search function supplied by HIV Database network. Result After successful amplification and sequencing of target genes for 165 samples, sequences of protease gene and reverse transcriptase gene of each sample were analyzed though BLAST respectively, the subtypes deduced by the two gene segments can be coincided well for all 165 samples. The subtyping result indicates that different subtype HIV-1 has various distribution characters in populations infected by different routes. Additionally, a case infected with ADHK recombinant HIV-1 is firstly reported in China. Conclusion TRUGENE HIV-1 Genotyping System could be successfully used for HIV-1 subtyping analysis.
