摘要
建立了2种红豆杉的离体胚培养和植株再生系统.结果表明,4℃保存和流水冲洗能够在一定程度上破除红豆杉胚的休眠,不同培养基对种胚培养的影响没有明显的差别,经过低温处理的种胚萌发率由41.7%提高到69%,成苗率由33.3%提高到44.8%;流水冲洗预处理对种胚萌发也有促进作用,处理2 d后种胚的萌发率和成苗率都有明显的提高.
The system of embryo culture in vitro and plant regeneration in two Taxus species are established. The dormancy of yew embryos are broken to some extent by means of 4℃ preservation and water leaching. The effects of different culture medium on the embryo culture are not obviously different. The germination rate of the seeds could be increased from 41.7% to 69% and the seedling rate be increased from 33.3% to 44.8% when the seeds are stored in low temperature. The water leaching treatment promotes the germination rate of the embryo. The germination rate and seedling rate are both increased distinctly after two days.
