摘要
以新鲜菠菜叶为原料,建立叶绿素a和其他有效成份的分离提纯方法。以微型实验为手段,采用柱层析法,以轻体碳酸镁为固定相,正已烷、甲苯、氯仿、石油醚—丙酮(50:3,v/v)、石油醚—丙酮—甲苯(50:3:3,v/v)为流动相,用梯度洗脱方式,紫外可见分光光度计检测。结果表明。从新鲜菠菜叶中分离出五种有效成分:胡萝卜素、脱镁叶绿素、叶黄素、玉米黄质和叶绿素a。
To establish the method of extraction chlorophyll a and other effective compositions from fresh spinach leaves by using column chromatography with gradient through minitype experiments. The Five compositions are separated from fresh spinach leaves by the column chromatography of gradient with the light magnesium carbonate as immobile phase and hexane ,toluene, chloroform, petroleum ether-acetone （50： 3, v/v）, petroleum ether-acetone-toluene （50： 3： 3,v/v/v） as mobile phase. The five compositions are detected as 13-carotene,chlorophyll b without magnesium ,lutein,xanzenthin and chlorophyll a by UV detection method. The method was simple and easy to operate,it is available to extract the chlorophyll a and other effective compositons from the fresh spinach leaves.
