摘要
采用常规石蜡切片法研究了东北龙胆不定根下断面新生不定根(再生根)的发生过程;在室外露地条件下进行了断根取药后植株再定植的最佳时间探索;用高效液相色谱法对一年生再生根的龙胆苦苷含量进行了测定.结果是,东北龙胆根下断面有很强的再生能力,再生根的发生中心为形成层,属于直接器官再生,实验条件下再生根发生过程约为20天;露地实验表明,再生根在生育期各阶段均可发生,生产上宜在花期进行;再生根龙胆苦苷含量高于其他部位,可以保证药材质量.采用此方法进行龙胆药材的生产可以缩短生产周期1-2年,值得试行、推广.
In this paper we studied the regenerate adventitious roots( regenerative roots) of occurrence process about excised section of adventitious roots of gentiana manshurica Kitag by using the normal paraffin method. We found the best time of the replantation after excising root and taking the medicine outdoors. The contents of gentiopicroside in the annual regenerative Foots were tested by HPLC. The results showed that the excised roots of gentiana manshurica Kitag have stronger regenerative ability, the occurrence center of the regeneration is cambium, which belongs to the direct organ regeneration. The regenerative process of adventitious roots needs about 20 days. The outdoor experiment indicated that the regenerative roots can grow at each stage of reproductive period, and the optimum time is florescence in production. The content of gentiopieroside in regenerative roots is higher than that in other parts, which can guarantee the quantity of medicine. This method deserves to try and popularize , which can shorten the productive period of gentiana manshuriea Kitag for 1 - 2 years.
