EGF对ER阴性乳腺癌细胞株MAPK通路及c-fos的活化作用 被引量:1

摘要 目的比较表皮生长因子(EGF)对ER(+)和ER(-)乳腺癌细胞M APK信号转导通路及原癌基因c-fos表达的活化作用,探讨ER(-)的乳腺癌细胞激素非依赖性生长的可能机制。方法EGF和M APK抑制剂(PD 98059)分别和(或)联合作用于ER(+)乳腺癌细胞株M CF-7和ER(-)乳腺癌细胞株M DA-M B-435s,W est-ern b lot技术检测两种细胞中磷酸化M APK蛋白(p-M APK)及c-fos蛋白的表达。结果EGF对ER阳性和阴性的两种细胞株p-M APK和c-fos蛋白的表达均有上调作用,但M DA-M B-435s细胞中两种蛋白的激活明显高于M CF-7细胞,且c-fos蛋白表达与p-M APK蛋白表达一致。结论EGF通过激活M APK信号转导通路,活化原癌基因c-fos来刺激细胞恶性增殖可能是引起ER(-)乳腺癌细胞的激素非依赖性生长的重要机制之一。
