摘要
目的观察异丙酚对内毒素血症大鼠血清TNF-α、IL-1β及IL-10作用的影响。方法24只雄性Wistar大鼠分为三组,内毒素组(L组)、异丙酚+内毒素组(P组)和对照组(C组)。L组股静脉注入内毒素(LPS)8mg/kg,后持续输注生理盐水1ml·kg-1·h-1;P组股静脉注入LPS后静脉注射异丙酚10mg/kg,随后以10mg·kg-1·h-1维持;C组静脉注入等量生理盐水。于注射LPS后60、120、180、240min取血,用酶联免疫吸附法(ELISA)测定血清中肿瘤坏死因子(TNF-α)、白细胞介素-1β(IL-1β)及白细胞介素-10(IL-10)含量。结果L组血清中TNF-α、IL-1β及IL-10浓度显著升高(P<0.01),P组各项指标均明显低于L组(P<0.01)。结论异丙酚可显著抑制内毒素血症时TNF-α、IL-1β及IL-10的水平,减轻内毒素导致的急性肺损伤。
Objective To investigate the effect ofpropofol on TNF-α, IL-1β and IL-10 ofendotoxic shock rat. Methods Twenty-four male Wistar rats were randomly assigned to three groups: Control group(Group C), Endotoxin group(Group L),and propofol plus Endotoxin group (Group P). Endotoxin with concentration of 10 mg/kg were injected into the rats of group L through femoral vein; The rats in group P had 20 mg/kg/h ofpropofol injected intravenously and then received 10 mg/kg/h of endotoxin; The rats m group C had the same amount of normal saline injected. The rats were killed 60, 120,180 and 240 minutes after the injection. The levels of TNF-α, IL-1β and IL-10 were measured by ELISA. Results In Group L, the concentrations of TNF-α, IL-1β and IL-10 rose significantly (P〈0.001). In Group P, all the concentrations were significantly lower than that of Group L (P〈0.001). Conclusion Propofol can significantly decrease the level of TNF-α, IL-1β and IL-10 in endotoxic shock, and then prevent acute lung injury caused by endotoxin.
