摘要
目的探讨Ca2+对人表皮角质形成细胞(keratinocyte,KC)中PAI-3表达的调节作用。方法利用免疫细胞化学、RT-PCR方法,检测高低Ca2+浓度作用下,培养的人表皮KC中PAI-3的表达变化。结果PAI-3 mRNA及蛋白质在高低Ca2+浓度下培养的人表皮KC中均有表达,并且高Ca2+浓度培养条件下较低Ca2+浓度下明显增强。PAI-3阳性染色在低Ca2+浓度培养下主要位于核膜周围,而在高Ca2+浓度培养条件下主要位于细胞质。结论Ca2+调节人表皮KC中PAI-3的表达,PAI-3可能具有调节KC分化的作用。
Objective To explore the regulation of plasminogen activator inhibitor-3 (PAI-3) on human keratinocytes (KC) by calcium. Methods The immunocytochemical method (ICC) and reverse transverse PCR (RT-PCR) were used to determine the expression of PAI-3 in human KCs cultured in low and high calcium medium. Results The PAI-3 mRNA and protein were discovered in low and high calcium medium and the expression of the transcript and protein levels in high calcium medium were much higher than in low calcium medium. The positive stain mainly localized in perinuclear membrane in low calcium medium, and in cytoplasm in high calcium medium. Conclusion Calcium regulates the expression of PAI-3 in human keratinocytes, and PAI-3 may have important role in the regulation of keratinocyte differentiation.
