瘦素对人L-02脂肪变肝细胞胆固醇流出的影响

Effects of Leptin on Cholesterol Effiux in L- 02 Fatty Liver Cells

目的 观察瘦素对人L-02脂肪变肝细胞胆固醇流出及小凹蛋白（cavdin-1）、三磷酸腺苷结合盒转运体A1（ASCAI）mRNA表达的影响，探讨瘦素在非酒精性脂肪性肝病发生、发展中的作用。方法 用20％的医用脂肪乳配制成10％的浓度，与10％的胎牛血清培养基共同培养细胞，造模时间24h，造模后分别用瘦素处理，浓度分别为10^-5、10^-6、10^-7mol／L，处理24h。每纽均行油红“O”染色。用RT—PCR测定cavelin—1、ABCA1mRNA表达。结果 油红“O”染色显示，10％的脂肪乳处理24h能成功制造肝细胞脂肪变性模型。RT—PCR显示肝细胞脂肪变性后cavelin-1及ABCA1mRNA表达增加。模型组加入瘦素处理后cavelin-1、ABCA1mRNA表达明显增加，且与瘦素浓度呈正相关。结论 10％的脂肪乳能导致肝细胞脂肪变性，肝细胞脂肪变性时，10^-7-10^-5mol／L浓度的瘦素能上调cavdin—1、ABCA1mRNA表达，促进细胞内胆固醇的逆转运。提示瘦素对治疗非酒精性脂肪肝病有一定作用。

Objective To study the chonge of cavelin - 1 and ABCA1 mRNA expression in normal and nonalcohofic induced fatty fiver cell and to explore the effect of leptin. Methods Made fatty fiver cell model with 10% medical fatty acid for 24 hours. The model groups were divided into treatment 1 group with 10^-7 mol/L leptins,the treatment 2 group with 10^-6 mol/L leptins, the treatment 3 group with 10^-5 mol/L leptins. Kaluobukao group was used as positive control group, and another group without any treatment was used as normal control. Each group was stained with red oil. Reverse trancriptase - polymerase chain reaction （RT- PCR） was applied for tile determination of cavelin - 1, ABCA1 mRNA. Results Red “O” of oil pigmentation showed that 10% medical fatty acid could make fatty fiver cell model for 24 hours. After L - 02 fiver cells were turned into fatty fiver cell, RT - PCR showed that cavelin - 1, ABCA1 mRNA＇ s expression increased obviously. When adding leptin to fatty fiver cell, the RT- PCR showed that cavelin - 1 and ABCA1 mRNA＇ s expression increased obviously than model group, and the variation was positive correlation with leptin＇ s concentration. Conclusion When liver L- 02 cell is turned into fatty fiver cell, leptin can restrain the constructive metabolism of triglyceride and advance its oxidation. Also leptin can advance reverse cholesterol transport of the cholesterol.
