家蚕核型多角体病毒碱性磷酸酶基因(alk-exo)与杆状病毒的分子进化分析 被引量:3

摘要 基于为杆状病毒分类提供新的依据,克隆了家蚕核型多角体病毒苏州株(BmNPV-SU)碱性核酸酶基因(alk-exo)及其侧翼区,并进行序列和进化分析。BLAST检索显示,BmNPV-SUalk-exo上游的部分ORF109序列仅在AcNPV、RoNPV中检索到同源序列;下游区域在AcNPV、RoNPV、SlNPV的基因组中检索到同源序列。序列分析表明,BmNPV-SU的alk-exo基因编码420个氨基酸残基,下游区域存在2个正向的重复。比对结果分析显示,在22种杆状病毒中,alk-exo基因间核苷酸序列和氨基酸序列的同源性不高,变异较大。同时,利用MEGA软件包中的UMEGA算法建立了基于alk-exo基因的分子进化系统树,表明通过杆状病毒的进化可以反映宿主昆虫的进化,杆状病毒与其宿主昆虫之间存在共进化关系。 Alkaline nuclease gene (a/k-exo) and its flanking region of Bombyx mori nucleopolyhedrovirus strain Suzhou (BmNPV-SU) was cloned, sequenced, and analyzed. According to the BLAST result, the homology of part sequence of ORF109 on the upstream of BmNPV-SU alk-exo gene had been found only in AcNPV and RoNPV, and the downstream region was homologous to AcNPV, RoNPV, and SINPV. Sequence analysis showed that BmNPV alk-exo gene encoded 420 amino acids and its downstream region contented two direct repeated sequence. Alignment between 22 baculoviruses showed that either nucleotide or amino acid sequence of alk-exo gene homogeneity was low, variation was high. Phylogenetic tree of alk-exo gene, which was constructed using UMEGA method in MEGA software, revealed coevolution of baculoviruses and their hosts. The evolution of hosts could be reflected by baculoviruses' evolution, which supplied new proof for virus taxonomy.
