摘要 目的观察羧甲基壳聚糖(CMC)对黑腹果蝇寿命的影响及其对D-半乳糖致亚急性衰老小鼠的抗衰老作用. 方法用含CMC(1%、2%、4%)的培养基喂养果蝇,观察其寿命;同时用不同剂量的CMC(100、200、400 mg/kg)对D-半乳糖模型小鼠灌胃,测定小鼠的学习记忆成绩、血清中超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量并分析胸腺系数和脾脏系数的变化情况. 结果 100、200 mg/kg CMC均能显著延长果蝇的平均寿命和最高寿命(P< 0.05~0.01);CMC还能显著改善衰老小鼠的学习记忆能力(P< 0.05~0.01),提高小鼠血清中SOD活性,减少 MDA含量,增大胸腺系数和脾脏系数(P< 0.01). 结论 CMC可能通过消除氧自由基、增强机体免疫功能而具有抗衰老的作用. Objective To observe the effect of carboxymethyl chitosan (CMC) on the lifespan of Drosophila melanogaster and D-galactose-induced aging in mice. Methods Drosophila melanogaster was fed with CMC ( 1% ,2% ,4% ), then the lifespan was observed. Different doses of CMC (100,200,400 mg/kg) were given to the old mice induced by D-galactose, then the ability of learning and memory, the activities of SOD, the level of MDA in serum, the thymus index and the spleen index were measured. Results Except the high dose group, the maximal lifespan and the mean lifespan were significantly prolonged (P〈0.01 or 0.05) ; the ability of learning and memory, the activities of SOD, the thymus index and spleen index were increased significantly (P〈0.01 or 0.05 ) and the level of MDA was decreased (P〈0.01). Conclusions CMC has anti-aging effect by cleaning the oxygen free radical and enhancing immune function.
