益生菌株对HT-29细胞的粘附及对致病菌的粘附抑制性能研究 被引量:9

摘要 目的研究培菲康微生态制剂中菌株的粘附性能及对致病菌的粘附抑制性能。方法以革兰氏染色法和酶联免疫吸附试验(ELISA)法评价3株益生菌分别对HT-29细胞和粘蛋白的粘附性能,与益生菌共孵育后裂解细胞活菌计数法检测益生菌抑制致病菌对HT-29细胞的粘附性能。结果嗜酸乳杆菌、长双歧杆菌和粪肠球菌的粘附指数分别为501.33±38.18,440.67±60.93,163.33±4.16,并可以分别使致病性大肠杆菌粘附率由6.7%下降到2.85%,0.214%,1.9%;使伤寒沙门氏菌粘附率由32.49%下降到16.95%,12.93%,20.33%。结论培菲康微生态制剂中3株益生菌均有不同程度的粘附性能以及抑制致病菌对肠上皮细胞粘附的作用,其中嗜酸乳杆菌的粘附性能最好,而长双歧杆菌对致病菌的粘附抑制作用最好。 Objective To investigate the adhesion capacity of three probiotics in Bifico and the adhesion inhibition of two bacterial pathogens to HT- 29 cells provide. Methods Gram staining and ELISA methods were used to evaluate the capacity of the adhesion of three probiotics to HT- 29 cells and mucin respectively. Plating count after incubating with probiotics and cell disrupting to detect the adhesion inhibition of two bacterial pathogens to HT- 29 cells by three probiotics. Results The adhesion indexes of LA, BL and EF were 501.33 ±38. 18,440. 67±60. 93, 163.33 ±4.16 respectively. After incubating with LA, BL and EF the adhesion ratio of EPEC was decreased from 6. 7% to 2. 85%, 0. 214% and 1.9% respectively and SL from 32. 49% to 16. 95% , 12.93% and 20. 33% respectively. Conclusion Three probiotics of Bifico can inhibit the adhesion of bacterial pathogens to HT- 29 cells. LA expresses the strongest adhesion to HT- 29 cells and BL expresses the best adhesion inhibition to HT- 29 cells by bacterial pathogens.
