摘要
目的研究建立葛根色谱指纹图谱,提供较完整的质量信息,更有效地监控质量.方法采用RP-HPLC,色谱柱:Shimpack C18(4.6 mm×250 mm,5μm);流动相:A-乙腈;B-水(0.05%醋酸),梯度洗脱,0~20min,90%~90%B;20~60min,90%~60%B;流速为1.0 mL·min-1,柱温为40℃,检测波长为254 nm,进行了葛根药材以及提取物指纹图谱研究.结果运用梯度洗脱法得到的色谱图各色谱峰分离较好,达到指纹图谱要求.结论采用该法可为控制葛根药材、提取物以及其制剂的内在质量提供依据.
OBJECTIVE To establish the fingerprints of Radix Puerariae and its extract for the quality control. METHODS The fingerprints of Radix Puerariae and its extract were established by HPLC.The chromalographic separation was perlermed on a Shim-pack C18 eolumn (4.6 mm× 250 mm,5 μm) .The mobile phase consisted of acetontrile and 0.05% acetic acid with gradient elution.The flow-rate was 1.0 mL·min^-1, the column temperature was al 40℃ and the UV absorbance detection was at 254 nm. RESULTS Under the selected chromatographic conditions, a good fingerprint of Radix Puerariae were obtained. CONCLUSION The quality of Radix Puerariae, its extract and formulation can bye controlled effectively by HPLC-UV fingerprint.
