摘要
目的研究高热量饲料诱发胰岛素抵抗(IR)动物模型的特征以及影响因素。方法两种性别的SD大鼠随机分为正常组和由高蔗糖、高脂肪组成的高热量饲料喂养的模型组,模型组大鼠于高热量饲料喂养前腹腔注射小剂量链脲霉素。用胰岛素耐量、糖耐量及糖异生等试验评估模型动物的胰岛素敏感性。结果给予高热量饲料6周可诱发实验动物产生以腹型肥胖、糖耐量异常、胰岛素耐量异常、糖异生能力增强、肝脏脂质沉积等为特征的IR动物模型。高热量饲料的加工方式以及实验动物的性别影响IR动物模型的形成。结论高热量饲料可诱发IR动物模型,但受饲料的加工方式及动物性别因素的影响。
OBJECTIVE To observe the charae, ters of high-calorie diet-rod insulin resistant (IR) animal model, and to explore the affecting factors of the model. METHODS Female and male SD rats were randomly divided into the normal-calorie diet-fed group and the highcalorie diet-fed group. The rats of the high-calorie diet-fed group were given a relatively low-dose streptozotocin (STZ) before the high-calorie diet feeding. The insulin sensitivity of animal models was quantified by insulin tolerance test, glucose tolerance test anti gluconeogenesis test, etc. RESULTS After 6 weeks feeding, the rats showed obvious abdomen obesity, impaired glucose tolerance, reduced insulin toleranee, enhanced gluconeogenesis with lots of lipids accumulated in liver. The insulin resistant symptoms appeared in the high-calorie diet-fed animals were also relevant to the artifactitious pattern of the feed, and the gender of experimental animals. CONCLUSION High-calorie diet can lead to IR animal model, which is effected by the artifactitious pattern of the feed, and the gender of experimental animal.
