青稞中β-1,3-葡聚糖酶的纯化及其抗血清的制备

摘要 萌发的青稞种子经醋酸钠缓冲液抽提,50% ̄85%硫酸铵分级沉淀,DEAE-Cellulose52、CM-SepharoseFastFlow离子交换层析和SephadexG-75分子筛层析,得到具活性的β-1,3-葡聚糖酶。SDS-PAGE检测显示分子量27kDa左右的主带(95%)。将纯化的β-1,3-葡聚糖酶对新西兰兔进行免疫制备抗血清,双向免疫扩散测定效价为1:64。免疫印迹分析表明纯化的β-1,3-葡聚糖酶免疫杂交带亦在27kDa处。
