摘要
用直接负染和免疫电镜方法从我国浙江省(杭州、奉化、温州等地)、上海、北京、福建、湖南等地的13种天南星科大田作物:芋(Colocasia esadenta)、魔芋(Amorphophallus sinesis)、药用植物:半夏(Pinellia ternata)、掌叶半夏(P. cordata)、盾叶半夏(P. pedatisecta)以及观赏植物:马蹄莲(Zantedeschia aethiopica)、海芋(Alocasia macrorrhiza)、象耳芋(Colocasia gigantea)、台果芋(Syngonium podophygum)、黄金葛(Scindapsus aureus)、广东万年青(Aglaonema modestum)、花叶芋(Caladium bicolar)、剑叶喜林芋(Philodendium oxycadium)上均检测到芋花叶病毒(Dasheen mosaicvirus, DMV)。在免疫吸附-免疫修饰电镜水平上,该病毒与DMV抗血清和PVY抗血清均有强阳性反应。在人工接种条件下,DMV各分离物均不侵染烟草等非天南星科供试植物,但接种天南星科指示植物Philodendron selloum、半夏和掌叶半夏均表现系统症状。受DMV自然感染的天南星科作物叶中,病毒粒子的平均长度为752nm,病毒粒子的最大分布范围为667~882nm。从感病植物叶组织超薄切片中观察到紧密聚集和分散的线状病毒粒子和典型的Potyvirus柱状内含体。内含体同时具有风轮状(Pinwheel)、卷筒状(Scroll-like)和片层状聚集体(Laminated aggregate)。
Dasheen mosaic virus( DMV) was detected from thirteen Araceae crops from Zhejiang province (Hangzhou, Lingbo, Fenghua and Wenzhou), Shanghai, Beijing, Fujian, Hunan province by using directly negative staining and ISEM Those crops are Colocasia esculenta, Amorphophallus sinesis as vegetable and food crops, Pinellia ternata, P. cordata, P. pedatisecta as Chineses medicienal crops, Zantedeschia aethiopica, Alocasia macrorrhiza, Colocasia gigantea, Syngonium podophyilum, S. podophyilum, Aglanonema modestum and Philodendron oxycadium as ornamental plants. DMV had strong positive reaction with both an antiserum of DMV and two antisera of PVY under imuno-specific electron microscope. Those DMV isolates from infected Araceae crops didn' t infect Nicotiana tobacum and other non-Araceae plants tested, but caused systemic symptoms when inoculated onto Araceae seedlings of Philodendron selloun, Pinelia ternata and P. cordata. The main length of DMV particles detected from natural infected crops is 752nm, and the main distribution of the particles is from 667 to 828nm. Tightly aggregated and loose distributed filamentous virus particles and cylindrical inclusion bodies were observed under ultra-thin sections of infected leaf tissue by DMV. The inclusion bodies consist with pinwheel, scroll-like structures and laminated aggregates simultaneously. The purified virus particles are characterized as typical potyvirus morphologically.The results indicate that dasheen mosaic virus is the main.pathogenic virus on Araceae crops in China Pillenia, Scindapsus and Syngonium are new hosts of DMV.
