摘要
在pH5.0的六次甲基四胺缓冲液中,罗丹明B能够与蛋白质相互作用形成复合物。该复合物使罗丹明B在-0.87V处的极谱还原峰峰电流下降,在实验选定的最佳条件下,该峰峰电流的下降值同牛血清白蛋白(BSA)的含量在2.5~30mg/L范围内呈线性关系,检出限为1.3mg/L。该法已应用于血清样品中蛋白质的测定,分析结果满意。
Rhodamine B can react with bovine serum albumin to produce an electrochemically non-active supermolecular Rhodamine B BSA in the buffer solution of (CH2)6 N4 (pH 5.0). Rhodamine B has welldefined second order derivative linear-sweep polarographic wave at 0. 87 V(vs. SCE). Adding BSA into the Rhodamine B solution resulted in the decrease of the peak current but not change of peak potential. The decrease of the peak current is proportional to BSA concentration from 2. 5 to 30 mg/L, the limit of the detection is 1.3 mg/L, and the correlation coefficient is 0. 996, this method can be applied to determine BSA concentration.
