摘要
目的:探讨槐定碱(sophoridine,SR)对大鼠大脑皮层、海马谷氨酸(Glu)和γ-氨基丁酸(GABA)免疫阳性神经细胞的影响及中枢作用机制。方法:采用免疫组织化学方法(SABC法)和显微图像分析技术检测SR对大鼠皮层、海马谷氨酸和γ-氨基丁酸免疫阳性神经细胞变化的影响。结果:侧脑室注射槐定碱0.2mg/只,大鼠皮层、海马Glu免疫阳性神经细胞数明显增加,GABA免疫反应阳性细胞数显著下降。结论:SR引起中枢神经系统Glu及GABA的平衡失调可能是导致中枢兴奋作用机制之一。
Objective: To study the effects of sophoridine on the positive cells Glu and GABA immunoreactions in cortex and hippocampus of rats and the mechanism of its central pharmacological effects. Method: Immunohistochemistry method and micrographic analysis technique were employed to monitor the effect of SR on the alterations of positive ceils Glu and GABA immunoreactions n cortex and hippocampus of rats. Result: SR administrated icv 0.2mg/rat surprisingly increased the number of positive ceils Glu immunoreactions but decreased the number of positive cells GABA immunoreactions. Conclusion: The expression imbalance of Glu and GABA in CNS caused by SR may be one of the mechanisms in which LSR leads the effects of excitation in CNS.
