摘要
本文以紫花苜蓿(Medicago sativaL.)为主要对象,建立了利用皂苷元特征显色测定苜蓿总皂苷含量的定量方法。以香草醛-高氯酸为显色剂,冰醋酸为溶剂,使皂苷元显色,用分光光度法在波长为560nm处测定苜蓿皂苷元的含量。以3-O-[β-D-glucopyranosyl]-28-O-[β-D-xylopyranosyl(1→4)-α-L-rhamnopyranosyl(1→2)-α-L-arabinopyranoside]medicagenate酸水解后的苷元为标准品建立的回归曲线高度显著,线性和稳定性可靠。
One major triterpene saponins has been isolated from roots of Medicago sativa L and has been identified as 3-O-[β-D-glucopyranosyl]-28-O-[β-D-xylopyranosyl(1→4)-α-L-rhamnopyranosyl(1→2)-α-L-arabinopyranoside] medicagenate . Its aglycone was prepared as standard for colorimctric determination of alfalfa saponins.The used colorants were vanillin reagents and measurements were made at λ=560 nm.The results showed that the method is sufficiently accurate for assays in series.
