摘要
在滇池水域中取藻样,过滤、接种在MDM培养基中,置于光照温室中培养,提取出叶绿素-a,然后测定在不同波长下的吸光度值,用叶绿素-a法计算公式计算出叶绿素-a含量.确定适宜的波长范围是用叶绿素-a法测定滇池叶绿素-a生长变化的关键.实验研究结果表明,滇池藻类的叶绿素-a吸收光谱有两个最强的收区:一个是在波长为400~460nm的蓝紫光部分,另一个是在波长为640~660nm的红光部分.在这两个波段下,可测定Fe^2+对滇池叶绿素-a生长的影响情况。
The water samples are taken from Dianchi Lake. After filtered, it is inoculated in MDM, then it is put in glasshouse, the chlorophyll-a of algae is abstracted, the absorbance values of chlorophyll a are measured at the various wavelength, the content of chlorophyll a could be calculated with a formula. Therefore, to determine the suitable scope of wavelength in chlorophyll-a method was a key factor. The experimental results show that absorbance values are strong in the wavelength scope of 400 ~ 460 nm and 640 ~ 660 nm. The case study is carried out by adding Fe^2+ into experiment sample for measurement effects of Fe^2+ on ehlorophyll-a in Dianchi Lake under these two wavelength scope.
