钙通道阻滞剂对大鼠肝移植冷保存再灌注损伤中细胞内钙的影响 被引量:2

The influence of calcium channel blockers on intracellular calcium levels of cold preservation-reperfusion injury during liver transplantation in rats

摘要 目的研究钙通道阻滞剂对减轻肝移植中冷保存再灌注下肝细胞的钙超载及保护肝脏的作用。方法64只W istar大鼠行原位肝移植,按灌洗保存液的不同成分分组:⑴对照组:高渗枸椽酸盐腺嘌呤液(HCA);⑵实验I组:HCA液+维拉帕米;⑶实验II组:HCA+硝苯地平;⑷实验III组:HCA+硫氮卓酮,测定冷保存再灌注后的大鼠肝细胞内钙和肝功能,并在光镜和电镜下观察肝脏的结构。结果细胞内钙及肝功能测定,各实验组与对照组相比较差异有统计学意义(P<0.05);组织学观察,各实验组损伤轻于对照组。结论钙通道阻滞剂对肝移植中冷保存再灌注损伤的肝脏有保护作用,且三类药物中维拉帕米最强,硝苯地平和硫氮卓酮次之。 Objective To investigate the influence of calcium channel blockers on intracellular calcium levels of cold preservationreperfusion injury during liver transplantation in rats, Methods According to the ingredient of perfusion and preservation solutions, the animals were divided into four groups : the control group with HCA solution, the experimental group Ⅰ with HCA solution plus Verapamil, the experimental group Ⅱ with HCA solution plus Nifedipine and experimental group Ⅲ with HCA solution plus Diltiazem. The intracellular calcium of the rat hepatocytes and liver function were measured and the liver structure was observed under light and electron microscopy, Results There were significant differences between the experimental groups and the control group in intracellular calcium concentration and liver function ( P 〈0. 05 ). The histological injury in the rats of experimental groups was obviously milder than that of the control group, Conclusion Calcium channel blockers have protective roles of cold preservation-reperfusion injury during liver transplantation in rats.
