三氧化二砷对卵巢癌细胞端粒酶活性及hTERT蛋白表达的影响 被引量:5

摘要 目的观察三氧化二砷(As2O3)对卵巢癌细胞株A2780端粒酶活性的影响。方法应用端粒酶多聚酶联反应-酶联免疫测定(PCR-ELISA)方法检测As2O3作用后卵巢癌细胞系A2780细胞端粒酶活性的变化并使用流式细胞仪检测hTERT蛋白表达。结果0.25μmol/L As2O3(24~72h)可抑制卵巢癌细胞系A2780的端粒酶活性及hTERT蛋白表达,抑制作用呈时间依赖性效应。结论As2O3可以通过降低hTERT蛋白表达进而抑制卵巢癌细胞的端粒酶活性。 Objective To investigate the effect of As2O3 on the telomerase activity and hTERT protein expression of A2780 cells. Methods The telomerase activity was detected by TRAP. hTERT protein expressions were detected by flowcytometry. Results The absorbency of the cells (A values) was used to represent the telomerase activity. At 48h and 72h, the A values of the A2780 cells treated with As2 O3 decreased to 0. 351± 0. 051 and 0. 238±0. 024, respectively, compared to the A value of 0. 492±0. 051 in the control cells without treatment ( P〈 0.05 ). The positive rates of hTERT protein expression in A2780cells treated with As203 at 48h and 72h were 77. 237% ± 2. 872% and 47. 767 % ± 1. 326 %, respectively, compared to the positive rate of 89. 513±3. 389% in the control cells without treatment( P 〈 0.05 ). Conclusions As2O3 downregulated the telomerase activity and hTERT protein expression in A2780 cells.
