摘要 目的探讨CD40配体激活对胃癌细胞生长、增殖和凋亡的影响。方法MTT法检测不同浓度sCD40L作用后AGS胃癌细胞株存活率,选定最佳干预浓度;流式细胞术检测sCD40L最佳干预浓度作用后胃癌细胞株AGS、BGC-823细胞周期和凋亡的变化。结果sCD40L的最佳干预浓度为2μg/ml;与胃癌细胞株AGS、BGC-823共育48 h,sCD40L可使高表达CD40的AGS细胞生长抑制,细胞周期阻滞在G1期,而对CD40低表达的BGC-823细胞生长无明显影响,并且AGS、BGC-823细胞均无明显的凋亡变化。结论sCD40L可明显抑制CD40高表达胃癌细胞的生长,其机制是使细胞周期阻滞。 Objective To observe the effects of CD40 ligand activations on the growth, proliferation and apoptosis of gastric cancer cell lines. Methods The growth and proliferation of gastric cancer cell lines with different CD40 ligand concentration were measured by MTT, then the best CD40 ligand activating concentration were selected. Cell cycle and apoptosis of AGS,BGC-823 gastric cancer cell lines activating with the best concentration were analyzed by flow cytometry. Results The best CD40 ligand activation concentration was 2 μg/ml. Inhibition of growth and blockage of cell cycle were observed in AGS ceils expressing high levels of CD40 after CD40 ligand activating 48 hours. But BGC-823 ceils expressing low levels of CD40 didn' t respond to it . All of cell lines showed no significant changes in apoptosis. Conclusion The growth of gastric cancer cells expressing high level of CD40 is suppressed by CD40 ligand activation . And the mechanism may be to block cell cycle.
