摘要
目的研究SD大鼠骨质疏松形成过程中肝脏Hepcidin的变化趋势。方法用70 mg/kg/d维甲酸连续灌胃两周建立大鼠骨质疏松模型,用RT-PCR法测定第3、第8、第13天肝脏Hepcidin基因表达。结果肝组织Hepcidin基因表达在模型第8、第13天出现变化,其在RT-PCR电泳中表达降低。结论模型组肝组织Hepcidin基因的表达降低对于骨质疏松形成过程可能存在直接或间接关系,Hepcidin可能成为骨质矿化异常诊断的新焦点,为进一步研究改变骨质疏松提供新的思路。
Objective To investigate the expression changes of the liver hepcidin gene in the formation process of SD male rats osteoporosis model. Methods SD male rats osteoporosis model was developed with tretinoin ig 70 mg × kg^-1 × d^-1 for two weeks and the expression of the liver hepcidin gene with RT-PCR was measured at the 3rd,8th,13th day. Results Expression of the liver hepcidin gene in SD rats osteoporosis model group was decreased at the 8th and 13th day. Conclusion The low expression of hepcidin in model rats liver may has direct or indirect relation with osteoporosis formation. Hepcidin investigation may become anew focal point in diagnose of bone mineralization abnormality and may make great progresses in studying osteoporosis.
