脂联素受体1基因在大鼠脂肪组织中的表达及其与肥胖易感性的关系 被引量:3

Expression of Adiponectin Receptor 1 mRNA in Adipose Tissue of Rats and Its Relationship to Obesity Susceptibility

摘要 目的探讨高脂高能量饮食(HFHCD)诱导下肥胖易感(OP)大鼠与肥胖抗性(OR)大鼠腹膜后脂肪组织脂联素受体1(AdipoR1)基因mRNA表达的差异及其与肥胖易感性的关系。方法雄性SD大鼠18只,以高脂高能量鼠料饲养2周后,按体质量增值大小分组,增重最多的6只为OP组,增重最少的6只为OR组。OP、OR大鼠继续饲以4周后,观察各大鼠体质量变化和腹膜后脂肪组织湿重,采用RT-PCR技术检测脂肪组织AdipoR1基因mRNA表达水平。结果1.HFHCD诱导下,OP组大鼠体质量、腹膜后脂肪组织湿重均明显高于OR组(t′=4.48 P<0.01);2.OP组大鼠腹膜后脂肪组织中AdipoR1 mRNA表达水平低于OR组,但两者差异无统计学意义(t=2.23 P>0.05);3.大鼠腹膜后脂肪组织湿重与AdipoR1 mRNA的表达水平存在显著负相关(r=0.735 P<0.05)。结论AdipoR1表达于脂肪组织。相同饮食条件下,个体发生肥胖的易感性是影响AdipoR1水平的重要因素。 Objective To observe the different expression of adiponectin receptor 1 ( AdipoR1 ) mRNA in retroparitoneal adipose tissue between obesity- prone(OP) rats and obesity- resistant(OR) rats on a high- fat and high- caloric diet(HFHCD) and its relation- ship with obesity susceptibility. Methods After 2 weeks on the HFHCD, 18 male Sprague - Dawley rats were separated on the basis of body weight gain into 2 groups. Six rats with the biggest body weight gain were designated as OP group and 6 rats with the smallest body weight gain were designated as OR group. The OP and OR animals received the HFHCD for 4 more weeks. The body weights were recorded at the time of prepareing model. The wet weights of retroperitoneal fat pad were measured when rats were killed. The levels of AdiponR1 mRNA expression in adipose tissue were detected by polymerase chain reaction(RT - PCR) technique. Results 1. After 6 weeks on the HFHCD, the OP rats body weights and wet weights of retroperitoneal fat pad were significantly heavier than those of the OR rats( t' = 4.48 P〈 0.01 ). 2. The expressions of AdipoR1 rnRNA in retroperitoneal adipose tissue of OP rats were lower than those of the OR rats( t' = 2.23 P 〉 0.05). 3. There were outstandingly negative correlations between wet weight of retroperitoneal fat pad and expressions of AdipoR1 mRNA. Conclusions AdipoR1 mRNA is expressed in adipose tissue. Under the same condition of the HFHCD, the expression of AdipoR1 mRNA in adipose tissue may be related to the different susceptibility to obesity.
