摘要 研究了人体血小板活化因子中1-O-十六烷基乙酰甘油磷酰胆碱与1-O-八烷基乙酰甘油磷酰胆碱的胶束电动毛细管色谱分离条件与间接紫外吸收检测条件。首次采用高效毛细管电泳法成功地分析了人体血小板活化因子中两种主要磷酰胆碱的含量。 O-Hexadecyl-and 1-O-octadecyl-sn-glycero-3-phosphorylcholine(C16-AGEPC and C18-AGEPC)in the platelet activating factor of human blood were analyzed by micellar electrokinetic capillary chromatography (MECC) with indirect ultraviolet absorption detection at 254 nm. The optimum running buffer for the separation was prepared by containing 50 mmol/L sodium dodecy sulphate-20 mmol/L potassium hydrogen phthalate-10 mmol/L borax-3mol/L urea(pH=6. 8). The separation was completed within 10 min,The detection limits of C16-AGEPC and C18-AGEPC were the same,i.e.60 ng/mL(k=3).The analytical precisions(n=6) were 2.8%~3.0%and 1.4%~1.7% for the determination with peak height and for the measurement of the migration time, respectively,The application of this method to the clinical samples was demonstrated.
