摘要
目的:观察陡脉冲电场对荷瘤裸鼠肿瘤生长的影响,分析其杀伤肿瘤细胞的作用机制。方法:建立卵巢癌荷瘤裸鼠模型,设对照组和陡脉冲治疗组,记录治疗后21天各荷瘤裸鼠肿瘤的体积和重量;光、电镜观察陡脉冲对肿瘤组织的病理学改变。结果:陡脉冲治疗组裸鼠肿瘤体积与重量明显低于对照组(P<0.01);陡脉冲对肿瘤组织有明显的破坏作用,治疗靶区内见广泛的凝固性坏死、血栓形成,电镜下出现典型的细胞凋亡改变。结论:陡脉冲通过对肿瘤细胞的杀伤、诱导细胞凋亡和血栓形成抑制肿瘤生长。
Objective : To observe the effect of energy contmllable steep pulses (ECSP) on the growth of ovarian tumor-bearing nude mice and to analyze its mechanisms to damage tumor cells.Methods:Ovarian tumor model was successfully achieved in nude mice and then be divided into the control group and experimental group. The volume and weight of tumors in two groups were recorded after exposed 21 days to ECSP. Tumor tissues were obtained and preserved for routine HE and transmission electron microscope (TEM) observation.Results:The volume and weight of tumors in ECSP group were signircantly smaller than the control group( P 〈 0.01 ). ECSP has showed effectively destructive action on tumor tissue,extensive coagulation necrosis and throrrbosis in blood vessel in targeting zone. Necrotic and apoptosis phenomenon were preserved under TEM. Conclusions: ECSP can inhibit effectively the growth of tumor through its certain damage effect to tumor cell, induced cell apoptcels and thrombosis.
