大黄虫丸对大鼠脑出血模型海马组织氨基酸含量的影响 被引量:10

Effect of Dahuang Zhechong Pill on Content of Amino Acids in Hippocampal Tissue of Ischemic Cerebral Hemorrhagic Rats

摘要 目的观察大黄丸对大鼠脑出血模型海马组织氨基酸含量的影响,探讨大黄虫丸对脑出血大鼠保护作用的可能机制。方法选择胶原酶加肝素联合注射法诱导脑出血大鼠模型,采用反相高效液相色谱荧光法观察大黄虫丸对大鼠脑组织海马中谷氨酸(glutamic acid,Glu)、天门冬氨酸(aspartic acid, Asp)及γ-氨基丁酸(γ-aminobutyric acid,GABA)的影响。结果脑出血模型组3天后海马的氨基酸含量(μmol/g)Glu(15.2926±4.2429)、Asp(3.6384±3.1021)和GABA(0.3859±0.1846)含量明显的高于正常组Glu(8.5040±1.7794)、Asp(1.2669±0.4695)和GABA(0.1770±0.0472)和假手术组Glu(8.0057±1.1227)、Asp(1.4986±0.4174)和GABA(0.1829±0.04665)(P<0.05),而中药大黄虫丸治疗组的Glu (9.0550±1.7195)、Asp(1.8085±0.6862)和GABA(0.1993±0.0424)含量明显降低于脑出血模型组(P< 0.05)。结论提示大黄虫丸能抑制兴奋性氨基酸(Glu、Asp)的释放,纠正兴奋性氨基酸(EAA)/抑制性氨基酸(IAA)失衡,从而达到减轻脑组织损害的作用。 Objective To observe the effect of Dahuang Zhechong Pill (DZP) on the content of amino acids in hippocampal tissue of model rats of ischemic cerebral hemorrhage (ICH). Methods Collagenase and heparin were injected into caudate nucleus to establish ICH rat model. The content of glutamic acid (Glu), aspartic acid (Asp) and 7-aminobutyric acid (GABA) in hippocampal tissue was detected by anti-phase high-performance liquid chromatographic fluorimetry to observe the effect of DZP on the content of amino acids in brain tissue. Results Three days after modeling, the content of Glu, Asp and GABA was significantly higher in the model group than those in the sham-operation and the normal control group (P〈0.05), for Glu 15.2926± 4. 2429 μmol/g vs 8.0057±1. 1227μmol/g and 8.5040±1.7794μmol/g, for Asp 3.6384±3. 1021 μmol/g vs 1.4986 ± 0.4174/xmol/g and 1.2669±0.4695μmol/g, and for GABA 0.3859 ±0. 1846μmol/g vs 0. 1829±0.04665μmol/g and 0. 1770 ±0.0472μmol/g. The content of Glu (9.0550 ± 1.7195 μmol/g), Asp (1.8085 ± 0.6862μmol/g) and GABA (0. 1993 ± 0.0424 μmol/g) in hippocampal tissue in the DZP group was reduced significantly, as compared with that in the model group (P〈0.05). Conclusion DZP could reduce the release of excitatory amino acids (EAA-Glu, ASP) to balance EAA/inhibitory amino acid (IAA), so as to relieve ICH-induced brain injury.
