细胞因子对人眼眶脂肪细胞分化过程中TSHr表达的影响 被引量:1

摘要 目的:探讨细胞因子对体外培养的人眼眶脂肪细胞分化过程中促甲状腺素受体(TSHr)表达的影响。方法:对8例人眼眶脂肪细胞进行分化培养,在培养基质中分别加入IL-2I、L-4I、L-6I、L-10和干扰素-γ(IFN-γ)等细胞因子,12 d收集细胞,通过RT-PCR观察TRHr的表达;放射免疫法检测TSH刺激后细胞内环-磷酸腺苷(cAMP)的含量;油红染色提取法检测细胞内脂肪含量。结果:IL-6I、L-4I、L-10I、L-2等细胞因子处理组的目的条带均强于未处理组(均P<0.01),而IFN-γ组明显弱于其他细胞因子处理组(P<0.01),与未处理组无明显差别(P>0.05)。经细胞因子处理后的培养的人眼眶脂肪细胞对TSH刺激2 h后cAMP的产量以IL-6组为最高,是未给予细胞因子处理组(对照组)的2倍多(P=0.000),IL-4和IL-10,处理组也均有不同程度的增高(P=0.045,P=0.034),IFN-γ处理组对TSH刺激后cAMP的产量则明显低于无细胞因子组(P=0.000),而IL-2处理组则与无细胞因子组的差异无统计学意义(P=0.057)。结论:IL-6、IL-4I、L-10等细胞因子可促进人眼眶脂肪细胞分化过程中TSHr的表达和增加细胞内的脂肪含量,IFN-γ可抑制人眼眶脂肪细胞分化过程中TSHr的表达和降低细胞内的脂肪含量。
