反相高效液相色谱法测定复方黄柏酊中小檗碱含量

Determination of Berberine in Compound Huangbai Tincture by RP-HPLC

目的建立复方黄柏酊中小檗碱的含量测定方法。方法用中性氧化铝吸附法制备样品,采用反相高效液相色谱法测定含量,色谱柱为C18柱(200mm×4.6mm,5μm),柱温为25℃,流动相为水-乙腈-磷酸二氢钾-十二烷基硫酸钠(50∶50∶0.3∶0.2),流速为1.0mL/min,检测波长为345nm。结果盐酸小檗碱浓度在50～250μg/mL范围内与峰面积线性关系良好,回归方程为A=46680.1C-154.0,r=0.9998(n=5),平均回收率为99.06%,RSD=0.19%。结论该方法灵敏、准确,重现性好,可作为复方黄柏酊中小檗碱的质量控制标准。

Objective, e To establish a method for identification of berberine in Compound Huangbai Tincture. Methods The samples were extracted by the aluminum oxide（neutral） column. The determination was performed by HPLC using C18 column（200 mm × 4. 6 mm, 5 μm）, column temperature： 25℃. The mobile phase consisted of water - acelonitrile - potassium dihydrogen phosphate - sodium laurylsulate（50 ： 50 ： 0. 3 ： 0. 2）. The UV detection wavelength was 354 nm and flow rate was 1.0 mL/min. Results The calibration curve of berberine hydro chloride was in good linearity over the range of 50 - 250 μg/mL, and regression equation was A = 46 680. 1 C - 154. 0（ r = 0. 999 8）. The mean recovery was 99. 06% （ RSD = 0. 19% ）. Conclusion The method is sensitive, accurate and reproducible. It can be used to control the quality of Compound Huangbai tincture.
