摘要
目的研究不同配比的当归芍药散主要成分的变化与药效的关系。方法采用高效液相色谱法测定当归芍药散中芍药苷和阿魏酸的含量变化;用避暗法和水迷宫法测定小鼠的学习记忆功能;用化学刺激法测定镇痛药效;以脑匀浆自发脂质过氧化中MDA的生成以及Fe2+诱导小鼠肝匀浆脂质过氧化水平升高,观察抗氧化作用。结果当归芍药散归芍比为1∶1.34时益智作用最好;归芍比为1∶5.4时镇痛作用最强;归芍比为3∶1时,抗氧化作用尤佳。结论随着方中当归芍药用量不同,芍药苷和阿魏酸的含量发生变化,这可能是其功效主治发生改变的主要因素。
AIM To study on the change of main components and correlative drug effect of different dose Dangguishaoyao Powder (DSP). METHOD A HPLC analysis method was established to determine the content of paeonitlorin and ferulic acid in Dangguishaoyan- San;the ability of learning and memory was measured by step-through test and water-maze test, the analgesic effect was studied by chemical stimulation method and the antioxidation effect was studied by mensuring MDA production in brain homogenate and MDA production in liver homogenate induced by Fe^2+ . RESULTS When angelica and peony was 1 : 1.34, the learning and memory effect of DSP was most obvious when angelica and peony was 1:5.4,the analgesic effect was most obvious; When angelica and peony was 3:1 :the antioxidation was effect most obvious. CONCLUSION The proportion of angelica and peony was changed, the content of Paeoniflorin and Ferulicacid in different dose of DSS was changed. This maybe the fundamental factor which changed the marked efficiency in treating.
