摘要
目的研究大肠癌中ICAM-1基因的表达,旨在探讨其在大肠癌局部进展中的作用。方法应用流式细胞术(flowcytometrytechniqueFCM)测定60例大肠癌和15例正常对照组结肠标本的ICAM-1蛋白表达量,蛋白表达量以荧光指数(fluorescentindexFI)表示,并将大肠癌根据临床病理分期、癌细胞分化程度、细胞学类型分组比较。其结果用统计软件包SPSS11.5进行统计学处理。结果大肠癌组ICAM-1基因蛋白的表达量明显高于正常对照组,P<0.01;转移组大肠癌ICAM-1基因蛋白的表达量明显高于未转移组,P<0.01;大肠癌中高分化腺癌与低分化腺癌ICAM-1基因蛋白FI值比较差异有显著性,P<0.01。结论ICAM-1基因的表达与大肠癌的发生和进展关系密切,值得临床注意。
Objective To study the expression of ICAM-1 to explore it's roles in the ontogenesis of carcinoma of large intestine. Methods ICAM-1 gene protein of 60 carcinomas of large intestine and 15 normal large intestine tissue specimen were measured using flow cytometry technique (FCM).The expression of protein was expressed by fluorescent index (FI). The samples in malignancy group were compared according to their clinical pathologic stage, degree of differentiation of carcinoma cell, and histological type. All the results were analyzed with the statistic software of SSPS 11.5. Results ①The FI of ICAM-1 (carcinoma of large intestine versus normal mucosae)were respectively 2.355±0.382, 1.591±0.407,P〈0.01; ②Duke's stage C+D versus Duke's stage A+B were respectively 2.591±0.231,1.939±0.234, P〈0.01; The differences were significant between the low differentiation adenocarcinoma and high differentiation adenocarcinoma groups in FI of ICAM-1 gene protein (2.564± 0.256,1.929±0.245,P〈0.01). Conclusion The occurrence of carcinoma of large intestine is related to high expression of ICAM-1; ICAM-1 is related to genesis, infiltration and metastasis of carcinoma of large intestine; The ICAM-1 gene may act as important indices of clinical diagnosis, treament and prognosis.
