摘要
通过三亲本杂交将携带豌豆根瘤菌吸氢基因的质粒pAL_(618)转移到台湾毛豆根瘤菌292-C中,经抗性筛选、质粒检测和吸氢活性测定,得到能稳定遗传的结合株292-C2和292-C3。在自生条件下结合株均表现较高的吸氢活性,而受体株292-C不吸氢,在共生条件下结合株表现为高吸氢,而受体株表现为放氢。
The cosmid pAL618 containing the hydrogen uptake genes of Rhizobiumleguminosarum was transferred into Rhizobium Japonicum(Glycine max meer 292-C)bytriparent mating system,Two transconjugants 292-C2 and 292-C3 were selected in Rhizo-bium mininal medium containing Ampicillin(25mg/L)and Tetraeycline(10mg/L).Theexpression of hydrogenase activities of transconjugants were studied.The transconju-gants 292-C2 and 292-C3 showed high hydrogenase activity both in free-living and symbi-otic state whiie the recipient showed no hydrogen uptake activity in free-living state andhydrogen evolution in symbiotic state.
