摘要
目的:研究高脂血症家兔的动脉粥样硬化斑块中基质金属蛋白酶-13的表达,为进一步探讨动脉粥样硬化发病机制提供形态学依据。方法:将24只健康雄性新西兰白兔随机分为两组:(1)空白对照组(正常组)8只,每天给予普通饲料;(2)阳性对照组(模型组)16只,每天给予普通饲料+1g胆固醇+15%猪油。16周末所有实验用兔麻醉处死,各组行主动脉粥样硬化病理形态学观察,包括大体形态学观察、光镜观察。根据形态学特点再分成稳定斑块和不稳定斑块,通过免疫组化对斑块内的病变进行定量分析。结果:S-P免疫组化可见模型组中基质金属蛋白酶-13在粥样斑块中平滑肌细胞及泡沫细胞中的细胞核上呈阳性表达,而正常血管壁表达阴性。在稳定性斑块与不稳定性斑块对比中发现,MMP-13在不稳定斑块中表达明显较稳定斑块要高。结论:MMP-13在AS不稳定斑块中较稳定斑块阳性表达明显,显示MMP-13与AS斑块的不稳定性有密切联系。
Objective:To investigate the expression of matrix metal propteinase -13 (MMP- 13)in carotid plaque and to provide the important information for the pathogenesis of atheroschrosis (AS). Methods:24 New Zealand white rabbits were randomly divided into 2 groups : normal group (n = 8), model group( n = 16). Normal group always received normal chow;model group always received normal chow ,5% cholesterol and 15% lard. At the end of the sixteenth week, all rabbits were killed to performe the pathologic examinations. In addition , the number of grum - positive cell of MMP - 13 and MMP - 2were analyzed by immunohistochemitry showing diseases with atherosclerotic lesion. Results: From this experiment,it thows many MMP - 13 - positive cells in the fibrous. None in normal group. Conclusion:MMP- 13 could express in atherosclerotic plaques in rabbits and has much reliance with AS.
