摘要
本文对以酪氨酸为底物,通过酪氨酸酶酶促反应合成黑色素(melanin),结果表明,该酶促反应系统的反应时间为3~4h,反应温度为40±2℃,pH值为7.8~8.8,通气量为10L/min,底物浓度为1.2g/t,酶活力为80000u/L时,酶促反应系统的反应效率、稳定性较高,底物酪氨酸转化率高。本实验所得黑色素溶解于反应溶液,该黑色素紫外扫描光谱显示,在200~320nm处有强吸收,吸收值随波长减小而升高。该黑色素合成方法时间短,工艺简单,成本低,酶制剂来源广泛,反应参数可控。
The synthesis of melanin by tyrosinase enzymatic reaction with tyrosine as substrate was studied in this paper.The resuits are showed as follows:the time of the enzymatic w.action system is from 3 to 4 horns,temperature is 40± 2℃ ,pH value is 7.8~8.8, air flow is 10 L/min concentration of substrate is 1.2 g/L and the enzymatic activity is 80000 u/L.The enzymatic reaction system has a high efficiency and stability with this way for melanin synthesis. The ratio of transformation from the substrate is high. The melanin is soluble in reaction solution, The ultraviolet visible absorption spectrum of the melanin showed the absorbing value reduces with the wavelength rises and absorb betterly in 200 ~320 nm.It has short time, simple technique and low cost, The enzyme preparation can be obtained widely and the reaction parametes can be controlled easily.
