摘要
研究了阳离子聚丙烯酰胺絮凝剂与硅藻土助滤剂配合施用对α-淀粉酶发酵液的絮凝处理工艺。结果表明:适宜的絮凝条件为pH 7.0-7.5,温度30-35℃,阳离子聚丙烯酰胺用量0.4-0.5 g/L,硅藻土用量20 g/L,此时菌体絮凝率(FR)可达95%以上。α-淀粉酶发酵液经絮凝处理后,过滤速率 (FV)为未处理的9.36倍,滤液澄清率(TR)在98%以上,α-淀粉酶活力回收率达92%以上。
Treatment process of cationic polyacrylamide and diatomaceous earth for the flocculation of the α-amylase fermented broth was studied. The result showed that the flocculation ratio (FR) was more than 95% under the optimum flocculation conditions, where the pH of 7.0-7.5, temperature of 30-35℃, flocculant concentration of 0.4-0.5 g/L and diatomaceous earth concentration of 20 g/L. In comparison with a zero flocculant control, the filtration velocity (FV) and filtrate transparency ratio (TR) of α-amylase broth treated with flocculant were 9.36 fold and more than 98%, respectively, and the recovery ratio of α-amylase activity was more than 92%.
