摘要
目的:建立稳定的吗啡依赖戒断后条件性位置厌恶(CPA)大鼠模型。方法:给予Sprague-Dawley(SD)大鼠吗啡10mg.kg-1,每天2次,腹腔注射(ip),连续6.5天,在吗啡注射第6天,予纳洛酮催瘾注射:0.3mg.kg-1,ip,1次,并与条件性位置反射训练箱搭配。结果:条件性搭配后,研究组试验鼠在伴药侧的时间显著性减少。结论:慢性吗啡注射纳洛酮催瘾戒断后CPA大鼠表现出明显厌恶动机,可作为研究阿片类物质戒断所致厌恶的生物学机制的载体。
Objective: To develop the reliable conditioned place aversion (CPA) rat model bv naloxone-precipitated withdrawal from morphine-dependence. Methods: Naloxone (0.3mg.kg-1 intraperitoneally) was used to precipitate withdrawal symptoms in rats with morphin dependence (6.5d morphine injection. 10mg.kg-1 .intraperitoneally twice per day). By pairing the precipitated withdrawal symptoms with the apparatus for place conditioning. CPA was developed in the rats. Results: During the CPA test session, the actual time of the rats in the MN group staying in the naloxone-paired compartment decreased significantly compared with that during the preconditioning session. Conclusion: The rats developed the CPA produced by naloxone-precipitated withdrawal from morphine dependence present obvious aversive motivation and would be used to study the biological mechanisms underlying the aversion due to opiates withdrawal.
