D-半乳糖对小鼠肝脏影响的流式细胞术分析

摘要 目的以流式细胞术探讨D-半乳糖不同剂量、不同时间对小鼠肝脏活性氧(ROS)水平及细胞凋亡的影响。方法SPF级25-30gKM种雄性小鼠按体重随机分组(n=6)。剂量.反应关系设阴性对照组、80、150和450mg/kg D-半乳糖剂量组;时间-反应关系研究设阴性对照组、腹腔注射80mg/kg半乳糖4、8和16周。实验结束后,取肝组织制备单细胞悬液,流式细胞术分析肝细胞ROS水平及细胞凋亡率。结果肝细胞ROS水平和细胞凋亡率的流式细胞术分析显示,随着D-半乳糖给予剂量的增加,各剂量组指标明显升高(P〈0.01);80mg/ks D-半乳糖连续给予8、16周后。指标亦明显增高(P〈0.01)。结论在一定给予剂量及给予时间范围内,D-半乳糖可明显增加实验动物肝细胞ROS水平,诱导肝细胞凋亡,ROS水平的高低与细胞凋亡的发生具有一致性的变化趋势。 Objective To explore the effects of D-galactose on reactive oxygen species(ROS) and apeptosis of hepatocytes in mice by Flow Cytometry. Methods 25 - 30 g SPF KunMing male mice were randomly grouped by weight( n = 6) .80,150,450 mg/kg of D-galactose dose groups were used in the dose-reaction relationship study,and 4,8,16 of 80 mg/kg D-galactose were used in the timereaction relationship study. Hepatocytes' ROS and apeptosis were measured by flow cytometry(FC). Results Hepatocytes' ROS level and apeptosis rate analysis of FC showed that the indice increased with the increase of D-galactose dose ( P 〈 0.01 ), and the two indice also increased after being given 80 mg/kg D-galactose for 8 and 16 W( P 〈 0.01 ). Conclusion D-galactose can significantly increase ROS level and induce apoptosis in mice hepatocytes within certain concentration and time, and ROS level has the same change with apeptosis.
