摘要 选择BCR/ABL cDNA 0.3kb的接合区序列,标记Biotin-14-dCTP,与慢性粒细胞白血病(CML)患者的细胞涂片原位杂交,链霉亲和素-碱性磷酸酶显色系统检测,建立了原位杂交检测BCR/ABL基因表达的方法。对3例非CML的血液病患者(对照组)、18例CML初诊和8例治疗后的CML患者骨髓进行检测。CML初诊患者17/18为阳性,1例阴性反应来自Ph染色体阴性型患者,CML治疗后患者BCR/ABL mRNA表达3/8为阳性,5/8为阴性。结合各例骨髓增生活跃程度,表明BCR/ABL基因表达水平可以反映治疗效果。 A 0. 3 kb cDNA probe was selected specifically for the BCR/ABL gene junctional region .and detected the expression of BCR/ABL gene in patients of chronic myeloid leukemia (CML) by biotin-streptavidin-alkalin phosphate in situ hybridization. Twenty-six cases of CML including 18 prior to treatment and 8 having received chemotherapy,and 3 cases of non-CML controls were studied. The expression of BCR/ABL gene was positive in the prior to treatment CML patients while negative in the non-CML group;negative results were obtained in most of the treated patients group. Myelosup-pression were observed in the bone marrow of these negative patients. These implied that the expression of BCR/ABL gene was related to the active proliferation of bone marrow cells.
