体外培养的人牙周膜细胞在机械力作用下骨保护因子mRNA的表达 被引量:2

摘要 目的 研究机械力对体外培养的人牙周膜细胞骨保护因子(osteoprotegerin,OPG)信使RNA(messenger RNA,mRNA)表达的影响。方法 应用细胞加力装置对体外培养的人牙周膜细胞施予间歇性牵张力48小时,原位杂交检测细胞加力前后OPGmRNA的表达。结果 机械力作用下人牙周膜细胞OPGmRNA表达下降。结论 正畸骨改建过程中,机械力引起牙周膜细胞0PG表达的变化。这一结论为进一步研究人牙周膜细胞在机械力作用下参与骨改建的机理打下基础。 Objective To study the expression of osteoprotegerin (OPG)at messenger RNA (mRNA) level in human periodontal ligament cells (hPDLCs)under mechanical force loading in vitro. Methods HPDLCs cultured in vitro were loaded with intermittent stretching force for 48 hours. In situ hybridization was used to indicate the expression of OPG at mRNA level in hPDLCs with and without force loading. Results The expression of OPG mRNA was down-regulated by mechanical force. Conclusion The regulation of OPG expression in hPDLCs by mechanical force contributes to the orthodontic alveolar bone remodeling. The conclusion does the groundwork for the study on bone remodeling mechanisms in orthodontic treatment.
