Effects of Endothelin-1 on the Proliferation and Differentiation of Human Periodontal Ligament Fibroblast Cells in Vitro

摘要 目的:探讨不同浓度内皮素-1(ET-1)对人牙周膜成纤维细胞(human periodontal ligament fibroblastcells,HPLFs)增殖和分化的影响,为进一步探讨正畸牙齿移动的生物学机制奠定基础。方法:采用HPLFs体外培养技术和四唑盐(MTT)显色分光光度法及生化检测法观察ET-1对人牙周膜成纤维细胞增殖和分化的影响。结果:与对照组相比,10-8mol/L组ET-1对HPLFs具有显著的促增殖作用(P<0.01),10-6mol/L组ET-1对HPLFs的增殖具有明显的抑制作用(P<0.01)。ET-1对HPLFs的分化没有影响。结论:ET-1为促HPLFs生长因子,但是对细胞分化不产生影响,可能在正畸过程中牙周组织的改建中发挥作用。 Objective: To investigate the effect of endothelin - 1 ( ET - 1 ) with different concentrations on the proliferation and differentiation of human periodontal ligament fibroblast cells (HPLFs). Methods: Using in vitro human periodontal ligament fibroblast culture technique, the proliferation and alkaline phosphatase (ALPase) activity of HPLFs were evaluated by thiazolyl blue tetrazolium bromide (MTT) spectrophotometric assay and enzyme kinetics methods. Results: Compared with the control group, ET- 1 with 10^-8 mol/l concentration could induce the proliferation of HPLFs (P 〈0.01 ). ET - 1 with 10^-6 mol/l concentration could restrain the proliferation of HPLFs ( P 〈0.01 ). ET - 1 with any concentration had no effect on the differentiation of HPLFs. Conclusion: Endothelin - 1 which was mitogen factor on human periodontal ligament fibroblast cells may play a role in the rebuilding of periodontal tissues during orthodontic treatment.
