摘要
目的:探讨谷氨酸受体2(GluR-2)在疼痛模型大鼠脊髓后角表达的定位、定量变化、年龄差别及其意义。方法:应用免疫组织化学方法检测GluR-2在脊髓后角的表达,并用图像分析系统和Western印迹法进行定量处理。结果:疼痛刺激后,中年、青年组大鼠刺激侧脊髓后角GluR-2阳性反应均有增加。老年组大鼠脊髓后角刺激侧GluR-2免疫反应变化不明显。且青年大鼠刺激侧比中年大鼠刺激侧的增加量多。Western印迹结果与免疫组化结果一致。结论:疼痛刺激可以上调脊髓后角GluR-2的表达,但这种上调因年龄不同而存在差异。
Objective: To investigate the expression and significance of glutamic acid receptor-2 in the spinal dorsal horn of different age rats following injection of formalin in hindpaw. Methods: The expression of glutamic acid receptor-2 in spinal cord was detected by immunohistochemical method and Western blot analysis. Computer-assisted image analysis system was used for quantitative analysis. Results: After formalin injection into the hindpaw, the expression of glutamic acid receptor-2 was significantly increased in the ipsilateral side of spinal dorsal horn of young and middle-aged rats, but not in old rats. The increment of glutamic acid receptor-2 immunoreaction in young rats was more than that in middle-aged rats. Conclusion: The inflammatory pain can increase the level of glutamic acid receptor-2 expression in the spinal dorsal horn, but the increase may be related with the response of different age rats to pain stimuli.
