自发性高血压大鼠肾和肺动脉神经肽Y神经分布的计量学研究 被引量:2

Metrological Study of Innervation Containing Neuropeptide Y in Renal and Pulmonary Arteries of Spontaneously Hypertensive Rats

摘要 目的:研究动脉壁含神经肽Y(NPY)神经纤维分布密度及特征与高血压时细小动脉构型重建之关系。方法:17周龄自发性高血压大鼠(Spontaneously hyperensive rats,SHR)和同龄正常血压大鼠(采用Wistar-kyoto,WKY)各15只,分别取其肾和肺,常规石蜡包埋切片,用免疫组织化学方法及全自动图像分析仪,对其各级动脉NPY神经纤维,进行定性、定量检测。结果:SHR动脉壁NPY抗体免疫反应阳性物主要分布在中外膜,且以细小动脉分布密度最高。在动脉分叉处形成簇状。SHR肾、肺动脉NPY纤维的体密度(22.65±8.52~1.98±0.69)和积分光密度(12.84±8.32~0.64±0.27)均明显高于正常血压大鼠(7.91±5.70~0.90±0.42;2.95±1.45~0.30±0.19),差异有显著意义(P<0.01;P<0.05)。结论:原发性高血压病时阻力性动脉收缩兴奋性增加及中膜肥厚的重要机制之一是与细小动脉壁含NPY神经分布过剩有关。 Objective:To study the relationship between the characteristics of perivaseular innervation containing neuropeptide Y (NPY) and structurally remodeled small arteries and arterioles with hypertension. Methods: Selected kidneys and lungs were removed from 17-week-old spontaneously hypertensive rats (SHR, n=15) and normatensive Wistar-Kyoto rats (WKY, n=15), regularly embedded in paraffin and cut sections. Immunohistoehemical technique and image analysis system were used to determining qualitatively and quantitatively the nerve fibres containing NPY of the ivarteries. Results: The nerve fibres of positive immunoreaction for anti-NPY occurred in the adventitia and media of the arterial walls, with higher density in the small arteries and arterioles in SHR. The nerve fibres formed clusters at the bifurcation of the arteries. The stereodensity (22.65±8.52-1.98±0.69μm^3/μm^3x±s) and integral luminous density(12.84±8.32-0.64±0.27x±s) of innervation containing NPY of the renal and pulmonary arteries in SHR were significantly higher than those in WKY (7.91±5.70-0.90±0.42;2.95±1.45-0.30±0.19). respectively with (p<0.01-0.05). Conclusion:One of the important mechanisms in the rise of contractive excitability and media hypertrophy of resistant arteries in hypertension is relation to hyperinnervation containing NPY of the small arteries and arterioles.
