枳椇子醋酸乙酯提取物对肝纤维化大鼠血清学指标及TGF-β_1的影响

Effect of the Ethyl Acetate Extract of Zhi Ju Zi on Serum Makers and the Expression of TGF-β_1 in Rats with Hepatic Fibrosis

目的:观察枳椇子醋酸乙酯提取物对实验性肝纤维化的作用?椒?采用四氯化碳进行造模,用放免法(radioimmunoassay,RIA)检测大鼠血清中透明质酸(hyaluronicacid,HA)和层粘蛋白(laminin,LN)的含量,用逆转录聚合酶链式反应(reversetranscriptionpolymerasechainreaction,RTPCR)检测肝组织中转化生长因子β1(transforminggrowthfactorβ1,TGFβ1)的表达量,并与秋水仙碱比较。结果:经过治疗后,给药组HA的含量明显下降(P<0.05),其LN的含量与秋水仙碱组比较无显著性差异(P<0.05),而给药组TGFβ1的表达明显下降(P<0.05)。结论:枳椇子醋酸乙酯部位提取物能够降低血清学指标并减少TGFβ1的表达。

Objective ： To study the effect of the Ethyl Acetate Extract of Zhi Ju Zi on hepatic fibrosis. Mehtods： Model of liver fi- brosis in rats was induced by CCI4. The level of hyaluronic acid（HA） and laminin（LN） was detected by RIN. The expression of TGF-β1 of the rat liver was detected by RT-PCR. Results： After treatment, the level of HA was apparently decreased（ P 〈 0. 05） ; the level of LN wsn＇ t apparently different with the Colchicin group（ P 〉 0. 05 ） ; The expression of TGF-β1, was apparently decreased （ P 〈 0. 05 ） . Conclusion ： The Ethyl Acetate Extract of Zhi Ju Zi can effectviely decrease the level of serum markers of hepatic fibrosis and the expression of TGF-β1.
